Le jeu reprend les principaux personnages jouables des précédents jeux Mario Party, tels que Mario, Princesse Peach, Luigi et Bowser. Il y a une liste de 20 personnages, dont quatre doivent être débloqués. Toad n'est pas jouable et joue le rôle d'hôte et de juge de la fête. Super Mario Party introduit plusieurs nouveaux personnages dans la série, comme Monty Mole, Goomba et Pom Pom, et réintroduit des personnages qui n'apparaissaient que dans les spin-offs sur console portable, à savoir Diddy Kong et Bowser Jr.###Multiplayer
Le jeu offre une grande variété d'options multijoueurs. Jusqu'à quatre joueurs peuvent s'affronter dans le mode Party principal, à tour de rôle. Dans le mode Partenaire, les joueurs s'affrontent en équipes dans la veine de Mario Party : Star Rush. Super Mario Party permet à deux joueurs d'utiliser une seule Switch, chaque joueur prenant un des Joy-Cons. Les joueurs peuvent également synchroniser leurs consoles Switch en local.
